Attacking and killing Priscilla is counted as a sin, but it can be absolved. If you keep her alive, you will have an area that will always allow you to be invaded in. Killing Gwyndolin will place a permanent sin upon you, and you can be constantly invaded by Blade of the Dark Moon members when human and online.
The fire keeper at the first bonfire in Anor Londo will become hostile after you have defeated Gwyndolin. Yes, you can fight Gwyndolin without killing Gwynevere. You just have to step through the fog door where you join the covenant. Gwynevere is a character in Dark Souls. She is the Princess of Sunlight and daughter of Lord Gwyn. The Gwynevere you see in game is an illusion created by Gwyndolin in order to manipulate the player.
